An important issue in the assassination of President John F. Kennedy (JFK) relates to the CIA's National Photographic Interpretation Center (NPIC) and what they saw on the night of November 22, 1963. This topic is covered by Douglas Horne who had been part of the Assassination Records Review Board (ARRB) in the 1990s.

This is what one of the two men who worked on the film on either 11/23/63 or 11/24/63 at NPIC said was his impression. His name is Homer McMahon and he was the supervisor of the color lab at NPIC (the sad irony here is RFK had them move into a new state of the art facility in 1/63, and this allowed them to handle color), and he said in interviews with the ARRB that he viewed the film about "10 times" while making 8x10 still color pictures (he of course was told which frames to make pictures of). As reported by Dr. Mantik he said his impression was "JFK was hit 6-8 times from three directions" (he could tell by the body movement and reactions of JFK), but the suspicious SS agent, Mr. "Smith", said NO, he was hit just THREE times from behind. This is telling because they had NOT worked out the SBT yet as we all know the WC claimed he was hit just twice. Douglas Horne has written an article on the NPIC goings on that weekend. The fact it was the CIA’s place alone makes one wonder. In this article Horne mentions another man who was working on the Zapruder film at the NPIC that weekend. He too believed he was working with the original, but his copy was different from McMahon’s!

Peter Janney conducted an exhaustive series of MP3-recorded telephonic interviews of Dino Brugioni throughout the late winter and spring of 2009 (seven interviews altogether, beginning on January 30th and ending on June 27th), [4] and the upshot was that without any doubt whatsoever, Mr. Brugioni presided over a distinctly different briefing board event at NPIC the weekend following the assassination, using a distinctly different Zapruder film. Mr. Brugioni, like Mr. McMahon, also presided over the making of enlargements — blowup prints — from individual frames of the Zapruder film, which were then mounted on briefing boards. But his work crew was entirely different than McMahon's; the numbers of enlargements made differed significantly; the number of briefing boards made was different; and the format of the briefing boards made at Brugioni's event was distinctly different. Most significantly, the format of the Zapruder film delivered at Brugioni's NPIC event was distinctly different from the format of the Zapruder film delivered at McMahon's NPIC event. Yet each man believed, without any doubt, that he was working with the original film. And the two events occurred only one day apart. (Ibid.)

So TWO teams were working on a film they thought was the original copy, but was it? I would suggest reading Douglas Horne’s article as it is very good and very detailed.
The man who took it and viewed it could NOT say it was the same film and that is the key part!
Warren Commission supporters, and some conspiracy-believers, cannot say for a fact the film was NOT altered, thus, there is plenty of reasonable doubt. The film still contains the key frames that show JFK was killed by a shot from the front though and this shows that the official conclusion is incorrect all by itself.
